 if( pressed Enter for first time?) {
     firstTime = Time.time;
 }
 if( pressed Enter for second time?) {
     secondTime = Time.time;

     difference = secondTime - firstTime;
 }

I would recommend using TimeSpan for formatting the string

 using System;

 TimeSpan timeSpan = TimeSpan.FromSeconds(time);
 string timeText = string.Format("{0:D2}:{1:D2}:{2:D2}", timeSpan.Hours, timeSpan.$$anonymous$$inutes, timeSpan.Seconds);

Try this- float t = 0f;

 void Update()
 {
       if(Input.GetKeyDown(KeyCode.KeypadEnter) || Input.GetKeyDown(KeyCode.Return))
       {
                float timeDiff = Time.time - t;
                t = Time.time;
                Debug.Log("Time after last pressed - "+ timeDiff +" Sec");
       }
 }
