Can we use substances textures on Unity?

Right now: no. This is a feature we are working on for a future release.

I have just upgraded to the free version of 3.4. I have substance textures that came with Autodesk 3ds Max 2012. I can load them as an asset, view them in the Project folder and modify the sliders. But I cannot apply them to any object I bring in. Do I need the pro version to use Substance?

No you should be able to use them, make sure you drag and drop the Substance $$anonymous$$aterial on your object (round icon), and not the Substance root(square icon).
