How does A* pathfinding algorithm work in unity 3d?
Does A* use navmesh? How does it know where the obstacles are?

The other way around, NavMesh's use A*, not A* using NavMesh.
I do believe that behind the scenes NavMesh's use an A* derivative algorithm to calculate least-cost paths, but that doesn't stop anyone from creating their own which may suit their particular situation better.
